Expression profile of HMGB1 protein in human breast cancer

Objective: To investigate the cytoplasmic expression of HMGB1 protein and HMGB1/TLR4 signaling pathway in the different stage breast cancer. Methods: Real-time PCR and western blot technology were conducted to detect the expression of TLR4, HMGB1, and RAGE gene in following samples: 13 stage I breast tissues, 22 stage II breast tissues, 17 stage III &IV breast tissues and the corresponding adjacent normal mammary tissues. Results: The mRNA level of HMGB1, RAGE and TLR4 was higher in the breast cancer than one of normal tissue (p<0.05). Cytoplasmic HMGB1 expression was higher in the stage I and II breast tissues than in normal tissues (p<0.05). However, cytoplasmic HMGB1 expression was not associated with histologic grade. Conclusion: HMGB1 was expressed in the cytoplasmic tissue of the early stage of breast cancer. The expression of HMGB1 gene in cytoplasmic breast cancer tissue can be used as one of parameters for determining early diagnosis and prognosis.
