Intrinsically disordered proteins (IDPs) are widespread in nature, and they were found to have important biological functions, which are associated with various human diseases. Therefore, the research of IDPs provides promising strategy for new drug design. However, the structural and dynamic properties of IDPs pose a challenge to the drug designs that directly target their disordered regions. This review summarizes the recent research progress on the drug designs for IDPs, including targeting of IDPs’ ordered domains, inhibiting the binding of IDPs to their ligands and directly targeting the functional disordered domains of IDPs. Moreover, the examples of IDPs as a potential drug target and the obstacles encountered currently are discussed in detail. At last, one successful example of drugs virtual screening for an IDP is described.
